Properties and Biology

BBa_K4467000 is a protein coding sequence comprising the small fungal effector protein, PstSCR1 (Dagvadorj et al., 2017). This fungal protein possesses the characteristics of a proteinaceous pathogen associated molecular pattern (PAMP) that has been shown to activate immunity in the non-host plant species N. benthamiana (Dagvadorj et al., 2017). With roles in suppressing host immunity, mediating uptake of nutrients and enabling parasitism, PstSCR1 is predicted to provoke an immune response in non-adapted hosts when secreted into the plant apoplast. Within the plant, PstSCR1 binds to either SERK3A or SERK3B, two proteins belonging to the plant's immune system. The presence of a signal peptide (not included in this aprt) facilitates the ability of PstSCR1 to pass through cellular membranes (Dagvadorj et al., 2017). The sequence has three conserved (Y/F/W)x(C) motifs, one being located at the N-terminus as seen in other wheat rust fungal effector candidates (Dagvadorj et al., 2017).

Usage

As part of the 2022 UNSW_Australia project to design a peptide based inhibitor for Puccinia sp., we used PstSCR1 as a potential fungal target. This protein was expressed in NEB TURBO and NEB T7 express E.coli cells.
